چکیده

Intestinal and skin microflora can influence several systemic health behaviors. In this study, we hypothesized that gut tissue microbiota are linked, control the microbiota. To test this, compared intestinal responses in mice to co-housed littermates with full-thickness incisions on back or K14Cre-targeted expression of HYAL-1 epidermis. The latter represent a skin-specific model hyaluronidase activity produced by wounding but without inflammatory stress response. Both interventions greatly exacerbated disease when were subsequently challenged oral Dextran sulfate sodium salt(DSS ) (60% vs 0% mortality, 11-fold induction IL6 survivors, p<.001). DSS response was dependent microbiome as Germ-free SPF given Vancomycin rescued from effects injury K14HYAL1. 16S rDNA sequencing feces confirmed altered showed relative decrease gram-positive bacteria including protective bifidobacterium. Mechanistically, could be explained results spatial transcriptomics using 10X Visium colon wounded K14HYAL1 had increased Mucin 2(Muc2) Antimicrobial Peptide Regenerating Family Member 3(Reg3). qPCR immunochemistry validated Muc2 Reg3b transverse colon. Furthermore, modeled addition 6.8kDa hyaluronan fragments epithelial cells (HT-29), showing these HA resulted 1.7 fold greater 2 Reg3a (p<.01) vitro. Since mucin Reg3 known regulate intestine, findings demonstrate how alters susceptibility disease.

Burn Injury Alters the Intestinal Microbiome and Increases Gut Permeability and Bacterial Translocation

Sepsis remains one of the leading causes of death in burn patients who survive the initial insult of injury. Disruption of the intestinal epithelial barrier has been shown after burn injury; this can lead to the translocation of bacteria or their products (e.g., endotoxin) from the intestinal lumen to the circulation, thereby increasing the risk for sepsis in immunocompromised individuals. Sinc...

Environmental Particulate Matter Induces Murine Intestinal Inflammatory Responses and Alters the Gut Microbiome

BACKGROUND Particulate matter (PM) is a key pollutant in ambient air that has been associated with negative health conditions in urban environments. The aim of this study was to examine the effects of orally administered PM on the gut microbiome and immune function under normal and inflammatory conditions. METHODS Wild-type 129/SvEv mice were gavaged with Ottawa urban PM10 (EHC-93) for 7-14 d...

Mucin glycan foraging in the human gut microbiome

The availability of host and dietary carbohydrates in the gastrointestinal (GI) tract plays a key role in shaping the structure-function of the microbiota. In particular, some gut bacteria have the ability to forage on glycans provided by the mucus layer covering the GI tract. The O-glycan structures present in mucin are diverse and complex, consisting predominantly of core 1-4 mucin-type O-gly...

Lactobacillus plantarum LP-Onlly alters the gut flora and attenuates colitis by inducing microbiome alteration in interleukin-10 knockout mice

The association between inflammatory bowel disease (IBD) and gut microbes has been widely investigated. Our previous study demonstrated that Lactobacillus plantarum LP‑Onlly (LP) applied as a probiotic altered the gut flora and attenuated colitis in interleukin (IL)‑10 knockout (IL‑10‑/‑) mice. In the present study, metagenome sequencing was performed to investigate the gut microbiome in IL‑10‑...

Gut Microbiome: Westernization and the Disappearance of Intestinal Diversity

The environment shapes our intestinal microbiome. By contrasting the gut microbiomes of African hunter-gatherer and European subjects, a new study reveals that urbanization is associated with a loss of microbial organisms and genes. What will be the consequences of the lost biodiversity in the sanitized, western-diet world?
